The top 8 reagents that need to be noticed in the laboratory
1. Acrylamide
Acrylamide is a moderately toxic substance. Can be absorbed through the skin and respiratory tract into the human body, therefore, in handling and use must wear protective equipment, such as antivirus clothing, respirator and gloves. Acrylamide harm is mainly caused by neurotoxicity, but also reproductive, developmental toxicity.(http://www.hnhbsj.com)
Any of the following may be classified as the observed object of chronic acrylamide poisoning:
A. Contact with acrylamide local skin appears sweaty, wet and cold, desquamate, erythema;
B. Limb numbness, stinging pain, lower extremity fatigue, lethargy and other symptoms;
c. Nerve-electromyography showed suspected neurogenic damage.
Treatment:
B vitamins, energy mixture, plus physical therapy, physiotherapy and symptomatic treatment. Support therapy should be strengthened at the same time.
2. NN- methylene bisacrylamide
NN- bisacrylamide is toxic and affects the central nervous system. Do not breathe in powder.
3.DTT dithiothrenol(http://www.hnhbsj.com)
DTT dithiothritol is a strong reductant that gives off a bad smell. May be inhaled, swallowed or absorbed by the skin and endanger health. When using solid or high concentration storage solution, wear gloves and goggles and operate in the ventilation cabinet.
4.TEMED
TEMED has strong neurotoxicity, prevent aspiration by mistake, operate quickly and seal when stored.
5. PMSF
Phenylmethyl sulfonyl fluoride [(PMSF), C7H7FO2S or C6H5CH2SO2F] is a highly toxic cholinesterase inhibitor. It is very destructive to the mucous membrane, eyes and skin of the respiratory tract. It can be fatal if inhaled, swallowed or absorbed by the skin. Wear suitable gloves and safety glasses and always use them in a chemically ventilated closet. In the case of contact, wash the eyes or skin with a large amount of water immediately. Contaminated overalls are discarded.
6. Chloroform (CHCl3)
Chloroform stimulates the skin, eyes, mucosa and respiratory tract. It is a carcinogen that can damage the liver and kidney. It is also volatile and avoids inhaling volatile gases. Wear suitable gloves and safety glasses at all times and always in a chemical ventilation closet.
7. Formaldehyde (HCOH)
Formaldehyde is toxic and volatile and is also a carcinogen. It is easily absorbed through the skin and stimulates and damages the eyes, mucosa and upper respiratory tract. Avoid inhaling volatile vapors. Wear the right gloves and safety glasses. Always operate in a chemical ventilation cabinet. Keep away from heat, sparks and open fires.
8.EB:EB (Ethidium bromide, ethidium bromide)
Ethidium bromide is a highly sensitive fluorescent dye for the observation of DNA. in agarose and polyacrylamide gels. Ethidium bromide is a strong mutagen with high carcinogenicity! It evaporates at 60-70 degrees (so it's best not to add it when the glue is too hot, or to add it to a liquid, 0.5ugml / ml, dyed for half an hour) (when EB is added too much, 20min can also be soaked in stained gels at room temperature to reduce background fluorescence caused by unbonded EB.
Purification of ethidium bromide solution: because ethidium bromide has certain toxicity, the solution containing EB should be purified and discarded after the experiment, in order to avoid environmental pollution and harm to human health.
(1) for solutions with EB content greater than 0.5mg/ml: (1) dilute EB solution to a concentration below 0.5 mg / ml; (2) add twice volume of 0.5mol/L KMnO4, then add equal amount of 25mol/L HCl, to mix, and put it at room temperature for a few hours; add twice volume of 2.5mol/L NaOH, to mix and discard.
(2) solution with EB content less than 0.5mg/ml: (1) add active carbon according to the quantity of 1mg/ml, mix gently from time to time, and leave at room temperature for 1 hour; filter with filter paper and live Carbon and filter paper are sealed and discarded.
